An alternative quantitative method and molecular genetics of amylose in Australian wheats

Jegasothy, Hemalatha (1999) An alternative quantitative method and molecular genetics of amylose in Australian wheats. PhD thesis, Victoria University of Technology.

Abstract

In this study, a simple and rapid electrophoretic procedure was developed to determine the amylose content in wheat starch. The experimental approach was based on agarose gels using a borate buffer and entailed optimisation of gel concentration, staining method, elecfrophoretic conditions and starch solubilisation conditions. Laser densitometry was used to quantify the amylose component.
